A:Yes, our product Universal-CE Phosphoramidite - (10-5000) should follow the same procedures as other the phosphoramidite reagents (Activation & coupling, capping, oxidation). Like other any phosphoramidites water should not be involved in the coupling of these reagents. Phosphoramidites are highly sensitive to water because it causes rapid hydrolysis destroying its ability to couple during oligonucleotide synthesis.

A:C3b is a fragment of C3.  It has no activity.

For C3b there is no functional assay in the CoA. However, we do an in-house functional assay wherein using SDS-PAGE analysis we determine the cleavage of C3b to iC3b by FI in the presence of cofactor FH.

We do evaluate all our C3b preparations in an in-house functional assay but we do not report it. We analyze Factor I cleavage of C3b to iC3b in the presence of the cofactor Factor H by SDS-PAGE under reduced conditions. C3b under reduced conditions shows two bands of about 101,000 daltons for the alpha prime chain (α’-chain) and 75,000 daltons for the beta chain. In the presence of cofactor Factor H, Factor I cleaves the alpha prime chain of C3b (101,000 daltons) yielding two fragments (63,000 and 39,000 Da). Thus, when C3b is converted to iC3b we see the disappearance of the alpha prime chain and the appearance of two bands @ 63,000 and 39,000 Da in addition to the beta chain @ 75,000 daltons. Functional analysis of C3b indicates that our C3b preparations are > 90% active.
